General description
Interleukin-10 is the archetypal anti-inflammatory cytokine that suppresses macrophage and dendritic cell activation, limits Th1 and Th17 responses, and maintains intestinal immune homeostasis. It is critical for preventing inflammatory bowel disease and autoimmune pathology. Recombinant Human IL-10 Protein (Y02501) is expressed from mammalian cells. It contains AA Ser 19 - Asn 178 (Accession # P22301), and is widely used in advanced cell culture, immunology research, and anti-inflammatory modeling.

Interleukin-10, also known as IL-10 or cytokine synthesis inhibitory factor, is a homodimeric cytokine that signals through a heterotetrameric receptor complex composed of IL-10R1 and IL-10R2 subunits, activating JAK1/TYK2-dependent STAT3 signaling. Unlike many cytokines that activate STAT3 to promote proliferation and survival, IL-10-STAT3 signaling in macrophages and dendritic cells selectively induces the expression of suppressor of cytokine signaling 3 (SOCS3) and other negative regulators that extinguish inflammatory gene expression programs. IL-10 is produced by regulatory T cells, B cells, macrophages, and intestinal epithelial cells, forming a critical feedback loop that prevents excessive immune activation and collateral tissue damage. Genetic deficiency in IL-10 or its receptor causes severe early-onset inflammatory bowel disease in humans, underscoring the non-redundant role of this cytokine in intestinal immune tolerance.
