General description

Interleukin-5 is a lineage-specific cytokine that regulates eosinophil production, differentiation, and survival. It is the principal driver of eosinophilia in allergic diseases and helminth infections, and serves as a therapeutic target in asthma and hypereosinophilic syndromes. Recombinant Human IL-5 Protein (Y08701) is expressed from mammalian cells. It contains AA Ile 20 - Arg 134 (Accession # P05113), and is widely used in advanced cell culture, immunology research, and allergy modeling.

Backgroud

Interleukin-5, also known as IL-5 or T cell replacing factor, is a disulfide-linked homodimeric cytokine that signals through a heterodimeric receptor composed of IL-5Rα and the common β-chain shared with IL-3 and GM-CSF receptors. This receptor configuration restricts IL-5 activity primarily to the eosinophil lineage, with IL-5Rα expression largely confined to eosinophils and basophils. IL-5 activates JAK2-dependent STAT5 signaling that promotes eosinophil precursor proliferation, terminal differentiation, maturation, and extended survival while preventing apoptosis. The cytokine also primes mature eosinophils for immunoglobulin-dependent degranulation and enhances their tissue migration capacity through upregulation of adhesion molecules and chemokine receptors. In allergic asthma, IL-5-driven eosinophilia contributes to airway hyperresponsiveness, mucus hypersecretion, and chronic tissue remodeling. Therapeutic monoclonal antibodies targeting IL-5 or its receptor have demonstrated clinical efficacy in severe eosinophilic asthma and eosinophilic granulomatosis with polyangiitis.
