Interleukin-4 is a Th2-polarizing cytokine that regulates humoral immunity, allergic responses, and alternative macrophage activation. It is essential for B cell class switching, eosinophil recruitment, and the suppression of Th1 inflammatory pathways. Recombinant Human IL-4 Protein (Y02201) is expressed from mammalian cells. It contains AA His 25 - Ser 153 (Accession # P05112), and is widely used in advanced cell culture, immunology research, and allergy modeling.
Th2 Cell Differentiation — Drives polarization of naïve CD4+ T cells toward IL-4-secreting Th2 effector populations.
B Cell Activation and Class Switching — Promotes IgE and IgG1 production in activated B lymphocytes.
M2 Macrophage Polarization — Induces alternative activation of macrophages toward tissue repair phenotypes.
Dendritic Cell Maturation — Supports the generation of mature antigen-presenting cells for vaccine research.
Serum-Free Culture Systems — Suitable for chemically defined and xeno-free cell culture media.

Interleukin-4, also known as IL-4 or B cell stimulatory factor-1, signals through the type I receptor complex composed of IL-4Rα and the common γ-chain, or the type II complex containing IL-4Rα and IL-13Rα1. This dual receptor usage explains the overlapping biological activities of IL-4 and IL-13 while preserving distinct signaling capacities in different cell types. IL-4 activates JAK1/JAK3-dependent STAT6 signaling that drives the master transcription factor GATA3, committing naïve T cells to the Th2 lineage and suppressing Th1 differentiation through downregulation of T-bet and STAT1 activity. In B cells, IL-4 promotes class switch recombination to IgE and IgG1 isotypes, underpinning allergic sensitization and humoral defense against helminth parasites. In macrophage biology, IL-4 induces alternative activation toward the M2 phenotype characterized by arginase-1 expression, mannose receptor upregulation, and tissue repair functions. EastMabBio's IL-4 is validated for Th2 polarization efficiency in primary human T cell cultures and supports reproducible dendritic cell maturation protocols for cancer vaccine development.
Q1: What is IL-4 mainly used for?
IL-4 is primarily used for Th2 cell differentiation, B cell class switching, M2 macrophage polarization, and dendritic cell maturation.
Q2: Is this IL-4 suitable for serum-free culture systems?
Yes. EastMabBio recombinant IL-4 is compatible with serum-free, xeno-free, and chemically defined media systems.
Q3: How should recombinant IL-4 be stored?
Lyophilized protein should be stored at -20°C or below. After reconstitution, aliquot and avoid repeated freeze-thaw cycles.
Q4: What shipping conditions are used?
Products are shipped on dry ice with validated cold-chain packaging to maintain stability during international transportation.
Q5: Do you provide QC documents?
Yes. COA, MSDS, TSE-BSE statements, endotoxin reports, bioactivity data, and additional QC documentation are available upon request.
