General description

Interleukin-21 is a pleiotropic cytokine produced by follicular helper T cells that regulates B cell affinity maturation, plasma cell differentiation, and the generation of stem cell-like memory T cells. It holds significant promise for enhancing the efficacy and durability of adoptive cell therapies. Recombinant Human IL-21 Protein (Y02901) is expressed from mammalian cells. It contains AA His 25 - Ser 162 (Accession # Q9HBE4), and is widely used in advanced cell culture, immunology research, and cell therapy manufacturing.

Stability and Reconstitution

Backgroud

Interleukin-21, also known as IL-21, is a member of the IL-2 cytokine family that signals through a receptor complex composed of the IL-21Rα chain and the common γ-chain shared with IL-2, IL-4, IL-7, IL-9, and IL-15. This receptor architecture places IL-21 at the intersection of multiple immune signaling networks while conferring unique biological properties. IL-21 activates JAK1/JAK3-dependent STAT3 signaling with weaker STAT5 activation compared to IL-2, creating a distinctive transcriptional program that favors effector differentiation over regulatory suppression. In T cell biology, IL-21 is unique in its ability to promote the generation of stem cell memory T cells that exhibit superior in vivo persistence, proliferative capacity, and anti-tumor efficacy compared to terminally differentiated effector cells. In NK cell biology, IL-21 enhances cytotoxicity and ADCC, making it valuable for combination strategies with therapeutic antibodies. In B cell biology, IL-21 drives plasma cell differentiation and immunoglobulin production while also promoting apoptosis of incompletely activated B cells to maintain selection stringency.
