General description
Tumor Necrosis Factor Alpha is a pleiotropic pro-inflammatory cytokine that regulates immune responses, cell survival, and apoptosis. It is a master regulator of inflammation and a therapeutic target in autoimmune diseases. Recombinant Human TNF-α Protein (Y04401) is expressed from mammalian cells. It contains AA Val 77 - Leu 233 (Accession # P01375), and is widely used in advanced cell culture, immunology research, and inflammatory disease modeling.
Application
· Macrophage and Monocyte Activation — Induces classical activation and pro-inflammatory cytokine production.
· NF-κB and Inflammation Signaling Research — Models TNF-α-driven inflammatory gene expression pathways.
· Apoptosis and Necroptosis Studies — Investigates cell death mechanisms through TNFR1 and TNFR2 signaling.
· Intestinal Organoid Barrier Dysfunction — Induces inflammatory phenotypes mimicking Crohn's disease and colitis.
· Serum-Free Culture Systems — Suitable for chemically defined and xeno-free cell culture media.

Backgroud
TNF-α is produced primarily by activated macrophages and signals through TNFR1 and TNFR2, activating NF-κB, MAPK, and death domain pathways. It is a central mediator of acute and chronic inflammation, with therapeutic antibodies (infliximab, adalimumab, etanercept) revolutionizing the treatment of rheumatoid arthritis, inflammatory bowel disease, and psoriasis. In cell culture, TNF-α is used to activate macrophages, model inflammatory signaling, study cell death pathways, and induce barrier dysfunction in intestinal organoids.
Q&A
Q1: What is TNF-α mainly used for?
TNF-α is primarily used for macrophage activation, inflammation signaling research, apoptosis studies, and intestinal organoid modeling.
Q2: Is this TNF-α suitable for serum-free culture systems?
Yes. EastMabBio recombinant TNF-α is compatible with serum-free, xeno-free, and chemically defined media systems.
Q3: How should recombinant TNF-α be stored?
Lyophilized protein should be stored at -20°C or below. After reconstitution, aliquot and avoid repeated freeze-thaw cycles.
Q4: What shipping conditions are used?
Products are shipped on dry ice with validated cold-chain packaging to maintain stability during international transportation.
Q5: Do you provide QC documents?
Yes. COA, MSDS, TSE-BSE statements, endotoxin reports, bioactivity data, and additional QC documentation are available upon request.

Recombinant Human TNFα (Cat. No. Y04401) was analyzed by SDS-PAGE.

Recombinant Human TNFα (Cat. No. Y04401) measured in a cytotoxicity assay using L-929 mouse fibroblast cells in the presence of the metabolic inhibitor actinomycin D. The ED50 for this effect is 5-60 pg/mL. The bioactivity of Y04401 was equivalent to other competing products.
