Interleukin-17A, also known as IL-17A or CTLA-8, is a disulfide-linked homodimeric cytokine produced primarily by Th17 cells, γδ T cells, and innate lymphoid cells. It signals through the IL-17RA/IL-17RC heterodimeric receptor complex, activating NF-κB and MAPK pathways through the adaptor protein Act1 to induce a characteristic transcriptional program of pro-inflammatory cytokines, chemokines, and matrix metalloproteinases. IL-17A is essential for protective immunity against extracellular bacteria and fungi, particularly at mucosal barriers, through recruitment and activation of neutrophils and enhancement of epithelial antimicrobial defenses.
