Validation Data
1. Serial‑passaging effect diagram
Throughout the continuous subculturing up to the 16th passage, the cells consistently maintained a favorable growth phenotype: clear nucleoli and tight intercellular junctions; the formed clonal aggregates exhibited a full, homogeneous morphology with well-defined borders; no cells exhibiting spontaneous differentiation were observed.
2. Alkaline phosphatase activity assay
After alkaline phosphatase staining, hPSCs exhibited a uniform blue-purple positive staining signal. This indicates that, even after multiple consecutive passages in culture, the cells still retain alkaline phosphatase activity.
3. Pluripotency marker identification
After multiple consecutive passages, the positive expression rates of the pluripotency markers OCT-4, SSEA4, TRA-1-60, and TRA-1-81 on P16 hPSCs all exceeded 98%, indicating that hPSCs maintained robust stem cell pluripotency even after continuous passage using the hPSC Culture Supplement (50×).
