General description
Bone Morphogenetic Protein 9 is the most potent endogenous inducer of osteogenic differentiation, acting through the ALK1 receptor to drive bone formation and vascular quiescence. It uniquely regulates both skeletal and vascular biology through distinct signaling mechanisms. Recombinant Human BMP-9 Protein (Y10801) is expressed from mammalian cells. It contains AA Gln 320 - Arg 429 (Accession # Q9UK05), and is widely used in advanced cell culture, vascular biology, and orthopedic research.

Stability and Reconstitution

Backgroud
Bone Morphogenetic Protein 9, also known as BMP-9 or GDF-2, is a member of the TGF-β superfamily that was originally identified as a circulating factor produced by the liver. BMP-9 is unique among BMPs in its high-affinity binding to the type I receptor ALK1 (ACVRL1), which is predominantly expressed on vascular endothelial cells, and its requirement for the co-receptor endoglin for optimal signaling. Through ALK1, BMP-9 maintains vascular quiescence and inhibits endothelial cell proliferation and migration, with genetic mutations in ALK1 or endoglin causing hereditary hemorrhagic telangiectasia (HHT), a vascular dysplasia characterized by arteriovenous malformations. In skeletal biology, BMP-9 is considered the most potent endogenous osteogenic factor, surpassing BMP-2 and BMP-7 in inducing osteogenic differentiation of mesenchymal stem cells through SMAD1/5/8-dependent activation of RUNX2 and Osterix. This dual role in vascular and skeletal biology makes BMP-9 particularly relevant for studying the coupling of angiogenesis and osteogenesis during bone repair. In liver biology, BMP-9 is produced by hepatocytes and regulates hepatic glucose metabolism and lipid homeostasis.
