General description
Interleukin-21 is a pleiotropic cytokine produced by follicular helper T cells that regulates B cell affinity maturation, plasma cell differentiation, and the generation of stem cell-like memory T cells. It holds significant promise for enhancing the efficacy and durability of adoptive cell therapies. Recombinant Human IL-21 Protein (Y02901) is expressed from mammalian cells. It contains AA His 25 - Ser 162 (Accession # Q9HBE4), and is widely used in advanced cell culture, immunology research, and cell therapy manufacturing.
Application
· Stem Cell Memory T Cell Generation — Promotes TSCM phenotype with enhanced in vivo persistence and anti-tumor efficacy.
· NK Cell Cytotoxicity Enhancement — Boosts antibody-dependent cellular cytotoxicity and target cell killing.
· B Cell and Plasma Cell Differentiation — Supports germinal center reactions and antibody production.
· CAR-T and TCR-T Cell Therapy Optimization — Improves therapeutic T cell product quality and durability.
· Serum-Free Culture Systems — Suitable for chemically defined and xeno-free cell culture media.

Backgroud
Interleukin-21, also known as IL-21, is a member of the IL-2 cytokine family that signals through a receptor complex composed of the IL-21Rα chain and the common γ-chain shared with IL-2, IL-4, IL-7, IL-9, and IL-15. This receptor architecture places IL-21 at the intersection of multiple immune signaling networks while conferring unique biological properties. IL-21 activates JAK1/JAK3-dependent STAT3 signaling with weaker STAT5 activation compared to IL-2, creating a distinctive transcriptional program that favors effector differentiation over regulatory suppression. In T cell biology, IL-21 is unique in its ability to promote the generation of stem cell memory T cells that exhibit superior in vivo persistence, proliferative capacity, and anti-tumor efficacy compared to terminally differentiated effector cells. In NK cell biology, IL-21 enhances cytotoxicity and ADCC, making it valuable for combination strategies with therapeutic antibodies. In B cell biology, IL-21 drives plasma cell differentiation and immunoglobulin production while also promoting apoptosis of incompletely activated B cells to maintain selection stringency. EastMabBio's IL-21 is validated for TSCM generation in primary human T cell cultures and supports the development of next-generation cell therapy products with enhanced therapeutic windows.
Q&A
Q1: What is IL-21 mainly used for?
IL-21 is primarily used for stem cell memory T cell generation, NK cell cytotoxicity enhancement, B cell differentiation, and CAR-T cell therapy optimization.
Q2: Is this IL-21 suitable for serum-free culture systems?
Yes. EastMabBio recombinant IL-21 is compatible with serum-free, xeno-free, and chemically defined media systems.
Q3: How should recombinant IL-21 be stored?
Lyophilized protein should be stored at -20°C or below. After reconstitution, aliquot and avoid repeated freeze-thaw cycles.
Q4: What shipping conditions are used?
Products are shipped on dry ice with validated cold-chain packaging to maintain stability during international transportation.
Q5: Do you provide QC documents?
Yes. COA, MSDS, TSE-BSE statements, endotoxin reports, bioactivity data, and additional QC documentation are available upon request.
SDS-PAGE

Recombinant Human IL-21 (Cat. No. Y02901) was analyzed by SDS-PAGE.
Bioactivity

Measured by its ability to enhance IFN-gamma secretion in NK-92 human natural killer lymphoma cells. The ED50 for this effect is ≤ 20 ng/mL. The bioactivity of recombinant Human IL21 (Cat. No. Y02901) is higher than the other competing product.
