General description
Noggin is a secreted BMP antagonist that binds BMP-2, BMP-4, and BMP-7 with high affinity, preventing receptor activation and maintaining stem cell populations in an undifferentiated state. It is an essential component of nearly all epithelial organoid culture protocols. Recombinant Human Noggin Protein (Y03301) is expressed from mammalian cells. It contains AA Gln 28 - Cys 232 (Accession # Q13253), and is widely used in advanced cell culture, organoid research, and stem cell biology.
Application
· Organoid Culture — Prevents spontaneous differentiation in intestinal, cerebral, and pancreatic organoid systems.
· Stem Cell Maintenance — Sustains pluripotency and prevents mesendodermal differentiation.
· Neural Induction — Promotes neural lineage specification by inhibiting BMP-mediated epidermal fate.
· Cartilage and Bone Research — Modulates BMP signaling in chondrogenic and osteogenic differentiation.
· Serum-Free Culture Systems — Suitable for chemically defined and xeno-free cell culture media.

Backgroud
Noggin, also known as NOG or SYM1, is a secreted glycoprotein originally identified by its ability to induce secondary axis formation in Xenopus embryos. It functions as a high-affinity binding protein for BMP-2, BMP-4, BMP-7, and GDF-5/6/7, forming inactive complexes that prevent receptor engagement. In organoid culture, Noggin is indispensable for maintaining the stem cell niche, as unopposed BMP signaling drives rapid differentiation and culture collapse. The protein is particularly sensitive to aggregation and precipitation, which can introduce unacceptable experimental variability. EastMabBio's Noggin is manufactured with optimized formulation conditions that minimize aggregate formation, ensuring consistent activity across extended culture periods and reducing the protein concentration required for effective BMP inhibition.
Q&A
Q1: What is Noggin mainly used for?
Noggin is primarily used for organoid culture, stem cell maintenance, neural induction, and BMP signaling modulation.
Q2: Is this Noggin suitable for serum-free culture systems?
Yes. EastMabBio recombinant Noggin is compatible with serum-free, xeno-free, and chemically defined media systems.
Q3: How should recombinant Noggin be stored?
Lyophilized protein should be stored at -20°C or below. After reconstitution, aliquot and avoid repeated freeze-thaw cycles.
Q4: What shipping conditions are used?
Products are shipped on dry ice with validated cold-chain packaging to maintain stability during international transportation.
Q5: Do you provide QC documents?
Yes. COA, MSDS, TSE-BSE statements, endotoxin reports, bioactivity data, and additional QC documentation are available upon request.

Recombinant Human Noggin (Cat. No. Y03301) was analyzed by SDS-PAGE.

Measured by its ability to inhibit 5.0 ng/mL of BMP-4 induced alkaline phosphatase production by ATDC-5 chondrocenic cells. The bioactivity of Recombinant Human Noggin (Cat. No. Y03301) is equivalent to competitor.
