R-Spondin 1, also known as RSPO1 or CRISTIN3, is a member of the R-spondin family of secreted proteins. Unlike Wnt ligands, R-Spondin 1 does not directly activate Frizzled receptors but instead potentiates Wnt signaling by binding to LGR receptors and inhibiting the E3 ubiquitin ligases RNF43 and ZNRF3, which otherwise target Frizzled receptors for degradation.
