General description
R-Spondin 1 is a secreted Wnt signaling potentiator that amplifies canonical Wnt pathway activity through LGR4/5/6 receptor engagement. It is the most critical and expensive component of standard organoid culture media, essential for stem cell self-renewal and long-term organoid propagation. Recombinant Human R-Spondin 1 Protein (Y03501) is expressed from mammalian cells. It contains AA Ser 21 - Arg 263 (Accession # Q2MKA7), and is widely used in advanced cell culture, organoid research, and regenerative medicine.

Stability and Reconstitution

Backgroud
R-Spondin 1, also known as RSPO1 or CRISTIN3, is a member of the R-spondin family of secreted proteins. Unlike Wnt ligands, R-Spondin 1 does not directly activate Frizzled receptors but instead potentiates Wnt signaling by binding to LGR receptors and inhibiting the E3 ubiquitin ligases RNF43 and ZNRF3, which otherwise target Frizzled receptors for degradation. This indirect mechanism of Wnt pathway amplification makes R-Spondin 1 remarkably potent at nanomolar concentrations. In organoid culture, R-Spondin 1 is absolutely required for the maintenance of LGR5+ intestinal stem cells and the long-term propagation of organoid lines. Its high cost has historically limited the scalability of organoid research, prompting EastMabBio to develop cost-effective bulk manufacturing processes that reduce the economic barrier to high-throughput screening and clinical applications without compromising biological activity.
