General description
Vascular Endothelial Growth Factor 165 is the predominant splice isoform of VEGF-A and the most potent angiogenic cytokine, driving blood vessel formation, vascular permeability, and endothelial cell survival. It is essential for developmental vasculogenesis, wound healing, and tumor angiogenesis. Recombinant Human VEGF165 Protein (Y04701) is expressed from mammalian cells. It contains AA Ala 27 - Arg 191 (Accession # P15692), and is widely used in advanced cell culture, angiogenesis research, and vascular biology.
Application
· Endothelial Cell Proliferation and Migration — Drives angiogenic responses in HUVEC and other endothelial cell types.
· Angiogenesis and Vascular Sprouting Assays — Models blood vessel formation in tube formation, spheroid, and aortic ring assays.
· Vascular Permeability Studies — Investigates VEGF-induced vascular leakage and edema formation.
· Tumor Angiogenesis and Anti-Angiogenic Therapy Research — Models pathological neovascularization and therapeutic intervention.
· Serum-Free Culture Systems — Suitable for chemically defined and xeno-free cell culture media.

Backgroud
Vascular Endothelial Growth Factor 165, also known as VEGF165 or VEGFA, is the most abundant and biologically active splice variant of the VEGF-A gene. VEGF165 contains both the heparin-binding domain encoded by exon 7 and the receptor-binding domains encoded by exons 3 and 4, enabling it to bind VEGFR2 with high affinity while also interacting with neuropilin-1 co-receptors and heparan sulfate proteoglycans to modulate signaling potency and tissue localization. VEGF165 signals through VEGFR2 to activate PLCγ-PKC, PI3K-AKT, and RAS-MAPK pathways, promoting endothelial cell proliferation, migration, survival, and vascular permeability. During embryonic development, VEGF165 is essential for vasculogenesis and angiogenesis, with even heterozygous deletion causing lethal vascular defects. In adult physiology, VEGF165 drives angiogenesis during wound healing, collateral circulation formation after ischemia, and female reproductive cycle vascular remodeling. Pathologically, VEGF165 is the principal driver of tumor angiogenesis, diabetic retinopathy, and age-related macular degeneration, making it a validated therapeutic target for bevacizumab, ranibizumab, aflibercept, and other anti-angiogenic agents. In cell culture, VEGF165 is indispensable for maintaining endothelial cell viability, establishing angiogenesis assays, and modeling vascular responses in tissue engineering constructs.
Q&A
Q1: What is VEGF165 mainly used for?
VEGF165 is primarily used for endothelial cell culture, angiogenesis assays, vascular permeability studies, and tumor angiogenesis research.
Q2: Is this VEGF165 suitable for serum-free culture systems?
Yes. EastMabBio recombinant VEGF165 is compatible with serum-free, xeno-free, and chemically defined media systems.
Q3: How should recombinant VEGF165 be stored?
Lyophilized protein should be stored at -20°C or below. After reconstitution, aliquot and avoid repeated freeze-thaw cycles.
Q4: What shipping conditions are used?
Products are shipped on dry ice with validated cold-chain packaging to maintain stability during international transportation.
Q5: Do you provide QC documents?
Yes. COA, MSDS, TSE-BSE statements, endotoxin reports, bioactivity data, and additional QC documentation are available upon request.

Recombinant Human VEGF165 (Cat. No. Y04701) was analyzed by SDS-PAGE.

Recombinant Human VEGF165 (Cat. No. Y04701) stimulates proliferation of HUVEC human umbilicvein endothelial cells. The ED50 for this effect is 0.2-10 ng/mL. Y04701 was higher than the other competing product.
