General description
Receptor Activator of Nuclear Factor Kappa-B Ligand is a TNF superfamily cytokine essential for osteoclast differentiation, lymph node development, and mammary gland morphogenesis. It is a therapeutic target in osteoporosis and bone metastasis. Recombinant Human RANKL Protein (Y07101) is expressed from mammalian cells. It contains AA Gly 64-Asp 245 (Accession # Q5T9Y4), and is widely used in advanced cell culture, bone biology, and cancer research.

Stability and Reconstitution

Backgroud
RANKL is a type II transmembrane protein of the TNF superfamily that signals through the RANK receptor, activating NF-κB, MAPK, and PLCγ pathways. It is produced by osteoblasts, stromal cells, and activated T cells. RANKL is indispensable for osteoclastogenesis, with genetic deletion causing severe osteopetrosis and absence of tooth eruption. Clinically, denosumab (anti-RANKL) is used to treat osteoporosis, giant cell tumor of bone, and skeletal-related events in cancer. In research, RANKL is the standard stimulus for generating osteoclasts from monocytes or bone marrow cultures.
