Wnt-3a is a canonical Wnt ligand that activates β-catenin-dependent signaling to drive stem cell self-renewal, proliferation, and lineage specification. The human Fc fusion format enhances protein stability and simplifies purification, enabling consistent activity in organoid and stem cell culture applications. Recombinant Human Wnt-3a-hFc Protein (Y04603) is expressed from mammalian cells. It contains AA Ser 19 - Lys 352 with a C-terminal linker GGGGGS and

Human lgG1 (Asp104-Lys330), and is widely used in advanced cell culture, organoid research, and developmental biology.

Backgroud

Wnt-3a, also known as WNT3A, is a member of the Wnt family of secreted glycoproteins that regulate cell fate decisions during embryonic development and adult tissue homeostasis. Canonical Wnt signaling stabilizes β-catenin, enabling its nuclear translocation and TCF/LEF-mediated transcription of target genes including MYC, CCND1, and LGR5. Native Wnt-3a is notoriously difficult to produce recombinantly due to lipid modification requirements and poor solubility. The hFc fusion format addresses these challenges by enhancing protein stability, improving purification efficiency, and extending half-life in culture medium. The Fc domain does not interfere with Wnt receptor binding but provides a convenient handle for protein detection and immobilization.
