The Human NK Cell Purification Cocktail cross-links non-target cells in human whole blood with multiple red blood cells (RBCs) to form immunorosettes. These rosettes increase the density of non-target cells, enabling them to sediment—along with free RBCs—during centrifugation with a density gradient medium. Target cells remain unlabeled by antibodies and are highly enriched, allowing for easy collection at the interface between the plasma layer and the density gradient medium.

01 Procedure

Based on the principle of density gradient centrifugation, simply add the K005 Human NK Cell Purification Cocktail to whole blood, incubate, and perform density gradient centrifugation. There is no need for expensive or complex magnetic or flow cytometry sorting equipment, saving time, effort, and budget.
Suitable for various sample types, including human peripheral blood and umbilical cord blood.

Q: What is the purity of NK cells after sorting?
A: One kit can process up to 50 mL of blood sample. If the sample volume is smaller (e.g., 10 mL), the experiment can still be performed by reconstituting the kit with 2 mL of sterile water and adding it at a concentration of 40 μL per mL of blood.
A: Purity may vary among donors, typically ranging from 40% to 75% after purification. If the initial purity is low, continuing standard culture protocols can increase purity to over 80% by Day 6 and over 90% by Day 10.
A: Depending on separation efficiency and blood sample quality:
A 10 mL blood sample typically yields 1–4 × 10⁶ cells
A 50 mL sample yields 0.5–2 × 10⁷ cells
For fresh blood samples, seed NK cells in a T25 flask with 9 mL of activation medium and 10% autologous plasma (≈1 mL), at a approximate density of 0.5–2 × 10⁶ cells/mL.
