Backgroud

Fibronectin, also known as FN1 or CIG, is a dimeric glycoprotein present in plasma and extracellular matrices. It contains multiple functional domains that bind integrins, collagen, fibrin, and heparan sulfate proteoglycans, enabling complex matrix assembly and cell-matrix communication. The central cell-binding domain containing the RGD motif interacts with α5β1 integrin to trigger focal adhesion formation and downstream FAK-Src signaling. In stem cell biology, fibronectin coating replaces undefined feeder layers, enabling fully defined, xeno-free culture conditions essential for clinical translation of iPSC-derived therapies. Fibronectin also plays critical roles in wound healing, embryonic morphogenesis, and tumor metastasis.
